<h4>Job summary</h4><p>The AGO is a ministerial department serving the Attorney General and the Solicitor General (the Law Officers). The Law Officers are the Government’s chief legal advisers. </p><p>The Law Officers make important decisions that affect public life. This role involves briefing (with or without supervision as appropriate) the Law Officers and the conduct of casework across the team’s responsibilities, including the unduly lenient sentencing scheme, consents to prosecute, contempt of court, family law cases, charity law cases and inquests. The successful candidate will conduct legal research across a wide range of issues, including constitutional law and devolution.<h4>Person specification</h4><ul><li>Candidates should have attention to detail, the ability to follow processes and ensure compliance with them, as well as the ability to analyse large volumes of information, to produce clear and concise summaries conveying only the key facts, and a reasoned and explicable recommendation. The AGO is a fast-paced department and candidates should be confident working at pace and advising on a wide range of legal issues.</li><li>It is desirable but not essential that candidates have experience of criminal law and/or public law.</li><li>The role primarily involves supporting the AGO’s work in relation to public interest casework, including, but not limited to, unduly lenient sentences, charity law, inquests and family law. This involves managing a busy caseload, interacting with external organisations, litigants in person and courts on behalf of the AGO, and drafting written submissions to the Law Officers. The postholder will conduct legal research across a range of issues including constitutional law and devolution and will support the AGO’s parliamentary and legislative work.</li><li>The postholder will need to work to short deadlines, briefing the Law Officers orally and in writing as necessary. Exceptional circumstances where SC may not be confirmed at time of starting will be dealt with on a case by case basis, but the continuation of post will rely on successful clearance being given. </p><p>The vacancy will be filled on loan or fixed-term appointment for an initial period of two years, with the possibility of an extension by mutual agreement. It is envisaged that the successful candidate will take up post in early September. If the incumbent is applying on temporary promotion, they will revert to their previous substantive grade and salary after the loan. There is the potential for permanent inward transfer, although this is by exception. Several criteria must be met concerning individual performance, business need and individual career development. A copy of AGO’s policy on secondment extensions and inward transfers can be made available upon request.</p><p>The transfer would be considered voluntary and so would not attract excess fares, travel costs or relocation costs.</p><p>Please note, due to the nature of loan agreements and the necessary release of staff into a new role, candidates will be required to discuss the loan and transfer with management at their home department prior to application. We will require confirmation that you have discussed this with management and they are content to release you into a new role, upon acceptance of any role. </p><p>Please note that any move across the Civil Service on or after October 2018 may have implications on an employee’s ability to carry on claiming childcare vouchers. You may however be eligible to claim tax free childcare. </p><p><strong>Salary</strong></p><p>Existing civil servants on level transfer will retain their current basic salary if it is within the AGO pay range. If your salary is higher than AGO maxima you will be placed on the maxima of the pay range and receive the difference as a mark-time allowance, if agreed by HR.</p><p>Where your salary is below AGO’s minima you will be raised to the minima of the pay range £36.758. Any reserved rights to London Weighting or associated London allowances, will be consolidated into basic salary before the uplift to AGO’s minima. Any other allowances will not be retained on moving to AGO. If this results in your salary exceeding the pay range maxima, you will retain the difference on a mark-time basis.</p><p>On promotion there will be an increase of 10% to your current basic salary, or move to the pay range minima, whichever is the highest. If you are in receipt of any reserved rights to London Weighting or associated London allowances, these will be consolidated after applying the 10% promotion calculation. After this calculation if your salary is below the minima of the pay range, you will be moved to the minima of the pay range. The Authorisations Team considers applications for the acquisition of communications data (CD) from law enforcement and public authorities. The team is located across two offices in Birmingham and Manchester.</p><p>Communications data is the information generated which relates mostly to telephone communication or internet activity. The data describes the communication but not the content of this.</p><p>Communications data is now used in 95% of serious and organised criminal prosecutions and has been used in every major counter terrorism investigation over the past decade. As such, the acquisition of CD must be independently authorised and that is the function of IPCO as an independent body.</p><h4>Job description</h4><p><strong>The role of an Authorising Officer</strong></p><p>Authorising Officers are responsible for assessing public authorities’ requests to acquire communications data in compliance with the Investigatory Powers Act 2016 and other relevant legislation.</p><p>Your role will be to independently assess the necessity and proportionality of applications received from public authorities. Applications are received from across the UK, and you will have the responsibility for the final decision as to authorise, decline or return applications.</p><p>The applications IPCO receive contain large amounts of information and need to be fully considered. Each application should be considered on its own merit, and you will make timely and objective decisions based on the facts available.</p><p>The role of Authorising Officer is vital as communications data can either support or disprove an alibi and can identify links that may exist between suspects and a crime scene. This is an additional allowance received alongside your basic salary, based on your attendance patterns for the year that compensates for working shifts. You will receive an annualised hours agreement, which is a written agreement between you and your manager, detailing the period the agreement runs for, your attendance requirements and your AHW percentages. This figure is currently circa 18%. This percentage calculation is assessed in line with the operational demands of the business and is subject to change and review. The studentship will offer a stipend, currently £22,780 per year and reviewed annually. </p><h4>Job description</h4><p><strong>About the Project</strong></p><p>Respiratory Syncytial Virus (RSV) causes significant disease burden, particularly in the very young and the very old. Recently there has been progress in preventing this virus, with the licensing of three vaccines and two monoclonal antibodies (mAbs Nirsevimab (Beyfortus) and Clesrovimab (MK-1654)). The success or failure of both antibodies and vaccines for RSV depends upon virus evolution and escape; this will particularly impact monoclonal antibodies. Another concern is that RSV immunisation may cause replacement of its ecological niche by the related winter virus human metapneumovirus (hMPV), itself a high priority amongst vaccine developers. An estimated 473,000 hMPV-associated hospitalisations occur globally, of which 185,000 were in high-income countries.</p><p>Hypothesis: Nanobodies that are cross-neutralising for RSV and hMPV can be generated.</p><p>The student will focus on the following topics:</p><ul><li>Generation of mRNA and proteins for pre-F from RSV and hMPV for immunisation. <ul><li>Based on the design of stabilised RSV pre-fusion F protein constructs and matching RNA vaccines developed by the Tregoning lab, the student will generate matching hMPV proteins and constructs.</li></ul></li><li>Immunisation of alpacas to recover nanobodies. <ul><li>Nanobody libraries will be generated from the immunised alpacas and appropriate selection strategies will be developed and employed to isolate cross-reactive nanobodies binding F protein of RSV and hMPV.</li></ul></li><li>Characterisation of nanobodies. <ul><li>Isolated nanobodies will be expressed in bacteria and characterised for binding (e.g., by ELISA or surface plasmon resonance). Their ability to neutralise RSV and hMPV will be assessed.</li></ul></li><li>Identification of escape mutants. <ul><li>Using classical virology techniques, RSV and hMPV viruses will be selected that escape neutralisation by the lead nanobodies. Escape mutants will be sequenced, thereby providing information about the epitopes recognised by the nanobodies.</li><li>The student will also model, using computational approaches, the identified amino acid changes on the respective F proteins, enhancing our knowledge about mutational tolerance of the F protein and the nanobody-binding interface.</li></ul></li><li>Testing of nanobodies in vivo as potential therapeutic agents. <ul><li>Using well established RSV challenge models, the student will explore protection against infection. The studentship will offer a yearly stipend, currently £22,780 and reviewed annually.</p><h4>Job description</h4><p><strong>About the Project</strong></p><p>Chimeric antigen receptor (CAR)-T cell therapy has shown great success in treating relapsed or refractory blood cancers, with several clinically approved treatments. However, current CAR-T therapies are associated with toxicological challenges, additionally involve complex, costly GMP manufacturing processes, and are associated with potential risks of introduction of viral components. The ErbB family of receptors, including epidermal growth factor receptor (EGFR), ErbB2, ErbB3, and ErbB4, play a crucial role in the development and progression of various cancers. Building up on the global success of the Covid-19 mRNA vaccine, this multi-disciplinary project will investigate mRNA-based in vivo CAR T-cell generation as a potential new approach for targeting T-cells for generation of efficient anti-tumour responses to gain an understanding of the immunotoxicological profile. The proposed project aims to leverage the mRNA platform to assess feasibility of in vivo reprogramming of circulating T-cells with a second- generation, pan ErbB-specific CAR (bearing the promiscuous ErbB ligand) for cancer immunotherapy, evaluating both anti-tumour efficacy and immunotoxicity. The in vivo approach will additionally employ targeting antibodies (Abs) to functionalise the mRNA lipid nanoparticles (LNPs) to facilitate uptake and targeted mRNA expression for in vivo therapeutic CAR T-cell induction. Recent studies indicate that the in vivo targeting approach produced CAR-T cells with similar anti-tumour activity to conventional CAR-T-cells and potential therapeutic benefit has been observed in disease conditions such as inflammatory cardiac fibrosis.</p><p>The successful candidate will acquire knowledge, expertise with training in a diverse range of laboratory techniques including LNP manufacture and functionalisation for selective targeting; in vitro, ex vivo and in vivo assays for therapeutic efficacy; and toxicological evaluation incorporating cellular, molecular biological and flowcytometry based assays.
