The student will be required to complete two 3-month rotations in the wider Agency, within their first year allowing them to gain unique insight into public health research.</p><p>The project will be supervised by Dr Deepa Rajagopal and Dr. Sandrine Vessillier at MHRA and Professor Vijay Chudasama at UCL. The studentship is anticipated to commence 1st October 2026. The studentship will offer a yearly stipend, currently £22,780 and reviewed annually.</p><h4>Job description</h4><p><strong>About the Project</strong></p><p>Chimeric antigen receptor (CAR)-T cell therapy has shown great success in treating relapsed or refractory blood cancers, with several clinically approved treatments. However, current CAR-T therapies are associated with toxicological challenges, additionally involve complex, costly GMP manufacturing processes, and are associated with potential risks of introduction of viral components. The ErbB family of receptors, including epidermal growth factor receptor (EGFR), ErbB2, ErbB3, and ErbB4, play a crucial role in the development and progression of various cancers. Building up on the global success of the Covid-19 mRNA vaccine, this multi-disciplinary project will investigate mRNA-based in vivo CAR T-cell generation as a potential new approach for targeting T-cells for generation of efficient anti-tumour responses to gain an understanding of the immunotoxicological profile. The proposed project aims to leverage the mRNA platform to assess feasibility of in vivo reprogramming of circulating T-cells with a second- generation, pan ErbB-specific CAR (bearing the promiscuous ErbB ligand) for cancer immunotherapy, evaluating both anti-tumour efficacy and immunotoxicity. The in vivo approach will additionally employ targeting antibodies (Abs) to functionalise the mRNA lipid nanoparticles (LNPs) to facilitate uptake and targeted mRNA expression for in vivo therapeutic CAR T-cell induction. Recent studies indicate that the in vivo targeting approach produced CAR-T cells with similar anti-tumour activity to conventional CAR-T-cells and potential therapeutic benefit has been observed in disease conditions such as inflammatory cardiac fibrosis.</p><p>The successful candidate will acquire knowledge, expertise with training in a diverse range of laboratory techniques including LNP manufacture and functionalisation for selective targeting; in vitro, ex vivo and in vivo assays for therapeutic efficacy; and toxicological evaluation incorporating cellular, molecular biological and flowcytometry based assays. Another concern is that RSV immunisation may cause replacement of its ecological niche by the related winter virus human metapneumovirus (hMPV), itself a high priority amongst vaccine developers. An estimated 473,000 hMPV-associated hospitalisations occur globally, of which 185,000 were in high-income countries.</p><p>Hypothesis: Nanobodies that are cross-neutralising for RSV and hMPV can be generated.</p><p>The student will focus on the following topics:</p><ul><li>Generation of mRNA and proteins for pre-F from RSV and hMPV for immunisation. <ul><li>Based on the design of stabilised RSV pre-fusion F protein constructs and matching RNA vaccines developed by the Tregoning lab, the student will generate matching hMPV proteins and constructs.</li></ul></li><li>Immunisation of alpacas to recover nanobodies. <ul><li>Nanobody libraries will be generated from the immunised alpacas and appropriate selection strategies will be developed and employed to isolate cross-reactive nanobodies binding F protein of RSV and hMPV.</li></ul></li><li>Characterisation of nanobodies. <ul><li>Isolated nanobodies will be expressed in bacteria and characterised for binding (e.g., by ELISA or surface plasmon resonance). Their ability to neutralise RSV and hMPV will be assessed.</li></ul></li><li>Identification of escape mutants. <ul><li>Using classical virology techniques, RSV and hMPV viruses will be selected that escape neutralisation by the lead nanobodies. Escape mutants will be sequenced, thereby providing information about the epitopes recognised by the nanobodies.</li><li>The student will also model, using computational approaches, the identified amino acid changes on the respective F proteins, enhancing our knowledge about mutational tolerance of the F protein and the nanobody-binding interface.</li></ul></li><li>Testing of nanobodies in vivo as potential therapeutic agents. <ul><li>Using well established RSV challenge models, the student will explore protection against infection. We’re looking for an experienced Principal Asset Manager to help us optimise Enfield’s diverse commercial estate and support the delivery of excellent services for our residents.<br>

The successful applicant will bolster the department’s role supporting Defra’s policy making and disease outbreak response activities.</p><p>In this role, you will be contributing to the production of advice for government policy and operational teams. This will involve building, maintaining or running models of disease systems that currently, or potentially could involve UK wildlife, and which can impact livestock and people. The post holder will be a member of the divisional training team in NPS or in HMPPS Wales. The post will be based within one of the NPS divisions or in Wales with some travel outside of that area</p><p>The post holder will report to the Divisional Training Manager/Deputy Divisional Training Manager. The training function is part of the HMPPS Learning &amp; Development Group.</p><p>The post holder works as part of the divisional training team to deliver probation qualifications at local level. This will include co-ordinating the needs of a group of learners and working with them individually and in groups both in the NPS and CRC.</p><p>Regular travel and some overnight stays are an integral part of this role. When required, PTAs will be expected to provide mutual assistance to other parts of HMPPS L&amp;D in order to meet the business need.</p><p>The role is a training function and the post holder will be required to apply their professional skills and specialist knowledge to ensure the effective delivery of probation qualifications and associated professional skills.</p><p>This post will not have line management responsibilities.</p><p><strong>Summary</strong></p><p>Co-ordinate and deliver all aspects of probation qualifications including brokering learning opportunities and monitoring the progress of learners based in NPS/HMPPS in England and Wales and in CRCs. Duties will include mentoring, assessment and coaching of learners in these organisations.</p><p>Assess VQ candidates in compliance with the requirements of the Awarding Body Design and deliver professional skills workshops</p><p>The post holder will be required to undertake quality assurance functions of learner work products and other quality functions dependant on business need. Relevant training will be provided.</p><p><strong>Responsibilities, Activities &amp; Duties</strong></p><p>The job holder will be required to carry out the following responsibilities, activities and duties:</p><p>• Participate in the selection of learners due to undertake probation qualifications</p><p>• Undertake the design, co-ordination and delivery of a range of training and learning opportunities to facilitate the learner’s achievement of probation qualifications.</p><p>• Identify individual learning needs, including reasonable adjustments and issues relating to learning style</p><p>• Complete and monitor a learning agreement for each learner in accordance with the Regulatory Framework</p><p>• Undertake regular developmental sessions with learners to identify ongoing needs and review progress on the qualification</p><p>• Identify performance issues and report these to the relevant line manager and to the Divisional Training Manager. Collaborate with the line manager to develop and an action plan to address issues.</p><p>• Provide written reports in respect of learner progress, including in relation to performance management.</p><p> • Proactively manage learner attendance at learning events and support managers in taking remedial action to manage sickness absence to achieve qualification completion.</p><p>• Provide coaching, mentoring, quality assurance and support the development of reflective practice in all aspects of work with learners and colleagues.</p><p>• Provide PTA cover in the absence of colleagues</p><p>• Attend all required training and assessment sessions and achieve the assessor award within 12 months of taking up post</p><p>• Use a variety of assessment methods with learners in line with the Awarding Body and sector assessment strategy, including written observations, performance feedback and digital recordings of professional discussions</p><p>• Maximise the use of IT and technology, including the use of the eportfolio system</p><p>• Attend standardisation meetings as required and liaise with Internal and External Quality Assurers to ensure the consistency and quality of assessments</p><p>• As and when required, undertake Internal Quality Assurance processes including chairing standardisation meetings; liaison with National Centre and divisional teams; attend Qualification Assurance Group as divisional representative and prepare for External Quality Assurance visits.</p><p>• Ensure assessment processes reflect best practice, are fair and consistent and meet the requirements of the Awarding Body</p><p>• Liaise with staff at the Higher Education provider to ensure learners’ progress towards achievement of academic awards and verify work based assignments, if required</p><p>• Develop and maintain effective working relationships with learners, colleagues and external agencies including CRCs</p><p>• Maintain probation professional knowledge by attending relevant training events and practical experience, as identified in the appraisal process.</p><p>• Travel regularly across the division/Wales and elsewhere, as required by the role</p><p>The duties/responsibilities listed above describe the post as it is at present and is not intended to be exhaustive.
